✱ Overview

What is NAD+?

Nicotinamide adenine dinucleotide (NAD+) is a coenzyme present in every cell. It shuttles electrons between chemical reactions, and it is consumed by enzymes that maintain DNA and regulate cellular stress responses.

NAD+ levels decline with age in several tissues, which has made it a focus of healthy-aging research. Most human research has studied oral precursors such as nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN), not injections.

✱ Mechanism

How it works

In mitochondria, NAD+ accepts electrons during glycolysis and the citric acid cycle, becoming NADH. NADH donates those electrons to the electron transport chain, which drives production of ATP — the cell's main energy currency.

NAD+ is also consumed by enzymes such as sirtuins and PARPs. PARPs help detect and repair DNA damage; sirtuins influence metabolism and stress responses. This is why NAD+ availability is studied in connection with aging.

Established clinical evidencePreliminary findingsExperimental hypothesis

01

Cellular energy & ATP

Established clinical evidence

NAD+ is indispensable to ATP production. This biochemistry is well established; it does not by itself show that injections raise energy levels.

02

Mitochondrial biology

Preliminary findings

Animal studies link restored NAD+ to improved mitochondrial function in aging tissue. Human data are early.

03

DNA repair pathways

Preliminary findings

PARP enzymes rely on NAD+ to repair DNA. Whether supplementation improves repair in healthy humans is not established.

04

Healthy aging research

Preliminary findings

Precursor trials show NAD+ levels can be raised in people, with mixed effects on clinical outcomes.

05

Cognitive research

Experimental hypothesis

Neuroprotection hypotheses are being studied in early trials, including in Parkinson's disease. No proven memory benefit.

✱ Evidence & safety

Read before you ask.

Evidence quality: Biological role: strong. Clinical benefit of injections: limited and largely unstudied in controlled trials.

Regulatory status: Compounded preparation · prescription required

Known safety concerns

  • Flushing, chest tightness or nausea during fast administration
  • Headache or abdominal cramping
  • Injection-site reactions
  • Limited long-term safety data for injectable use

Contraindications & cautions

  • Pregnancy or breastfeeding without clinician guidance
  • Active cancer — discuss with an oncologist, as NAD+ metabolism is studied in tumor biology

This information is educational and incomplete. Always seek advice from a qualified medical professional.

✱ FAQ

NAD+ questions

No. NAD+ is an active area of aging research, but there is no evidence that injections reverse aging.

Direct comparisons are limited. Most published human studies used oral NR or NMN.

These are research questions, not established benefits. Discuss persistent fatigue with a clinician, as it can have treatable causes.
